Why PRP Preparation Methods Matter: Tube Systems, Centrifugation and Platelet Concentration
Are all PRPs the same? Closed vs open systems, single vs double spin, platelet concentration, leukocyte content and activation, and how they affect results.
Not all "PRP" is the same. The system, centrifuge settings and concentration shape the plasma you receive.
| Variable | Why it matters |
|---|---|
| Blood volume | Determines PRP volume |
| Tube and system | Sterility and separation quality |
| Spin speed and time | Separation and platelet preservation |
| Single / double spin | Concentration level |
| Platelet concentration | Growth-factor density |
| Leukocytes | Possible inflammation and comfort effects |
| Residual red cells | May affect post-treatment tenderness |
| Activation | Timing of growth-factor release |
Closed systems keep everything sterile from draw to syringe, reducing contamination; open systems involve manual transfers and need meticulous technique.
Licensed, single-use closed systems are generally preferred.
| Method | Description |
|---|---|
| Single spin | Faster; concentration may be lower |
| Double spin | Plasma spun again; higher concentration possible |
Both aim to concentrate platelets without damaging them; excessive speed or time can impair function.
More isn't always better: concentration is often expressed as several times baseline; views differ on the ideal range for hair; too low may underperform, but extra benefit from very high concentrations is unproven.
| Type | Feature |
|---|---|
| Leukocyte-poor | Less inflammatory; common for hair |
| Leukocyte-rich | Used in some orthopaedic settings |
Activation: some protocols activate with calcium chloride, others rely on natural activation in tissue; both are used.
Ask your clinic: Which system — is it closed? Are tubes single-use and licensed? How much blood and PRP? What spin protocol? Who performs it?
Fatty meals can cloud plasma, so eat lightly beforehand.
At our Istanbul clinic PRP is prepared with single-use closed systems and a standard spin protocol, used only for you in the same session; clients from Yalova, Bursa, Kocaeli and Gebze can ask about our system at consultation.
